Influence of Adipose-Derived Adult Stem Cells on the Epidermis of Living Skin Equivalents

Abstract
There has been no ideal artificial skin model which can be used for replacing human skin or animal. But, the need of artificial skin is increasing for various purposes such as experimental model or clinical application. In this study, living skin equivalents(LSE) were reconstructed using adipose-derived adult stem cells(ADAS) containing collagen matrix instead of fibroblast(Fb) containing collagen matrix. ADAS were isolated from lipo-aspirated fat tissues and its phenotype was confirmed by cell surface markers of mesenchymal stem cell. Both ADAS-LSE and Fb-LSE were compared by using immunohistochemical staining and confocal microscopic examination. Also, human cytokine antibody array was done to analyze differences between Fb and ADAS. We were able to recognize that ADAS-LSE showed no parakeratosis and more complete differentiation. Furthermore, PCNA(-)/p63(+) keratinocytes was observed in basal layer. It may be suggested that these PCNA(-)/p63(+) cells can be stem cells that are not observed in the hyper-proliferative status of skin like Fb-LSE. Further investigation is needed to find out the mechanisms that can mediate these results. But, basic fibroblast growth factor(bFGF) and hepatocyte growth factor(HGF), which are expressed much more in ADAS maybe involved.
